产壳聚糖酶菌株的单因素及复合条件诱变育种

         

摘要

In this article,the Chitosanase M2 was used as original strata,the spore suspenslon oi whicn was re- duced by UV mutation, NANO: mutation and the compound treatmeat (UV + NaNO2) respectively to the vitali- ty of the Chitosanase of the population is for the purpose of A high-yielding mutant was obtained from a lot of mutants. The strain' s pullulanase activity reached 8. 135 U/mL. A 1. 381 increased than the original strain. Based on comparative analysis validation, composite mutation approach is a better way of looking for high ac- tivity of mutant strains. And a single factor in the ultraviolet mutation method is simple and instant, the en- zymes Chitosanase activity increased by 25.15% for complex mutation than the ultraviolet mutation, which is 32.82% higher than the nitric acid sodium mutation of the population of the enzyme activity.%以产壳聚糖酶菌株M2为出发菌株,分别采用紫外线诱变、亚硝酸钠诱变及两者的复合诱变方式对其孢子悬液进行诱变处理,以获得高产壳聚糖酶的突变菌株为目的。结果表明,突变株的酶活力高达8.135 U/mL,比原始出发菌株提高了1.381倍。经比较分析验证,复合诱变的方法是寻找高活力突变菌株的较好方法,而单因素的紫外诱变方法也具有简单快捷的优势,复合诱变比紫外线诱变后的产壳聚糖酶菌株酶活力提高了25.15%,比亚硝酸钠诱变后的菌株酶活力提高了32.82%。
