hBMP-7基因转染大鼠骨髓基质细胞促进骨质疏松大鼠牙周组织再生的实验研究

         

摘要

Objective This study is to assess the potential effects of bone marrow stromal cells (BMSCs) transfected by human bone morphogenetic protein-7 (hBMP-7) gene in periodontal regeneration of osteoporosis Sprague Dawley rats. Methods Osteoporosis models of the female Sprague Dawley rats were induced by ovariectomy. BMSCs from osteoporosis rats and sham operation rats were transfected by recombinant AD5-hBMP7-EGFP virus. BMSCs with different treatments were seeded on the collagen membranes. The membrane-cell complex and blank membrane were implanted into the acute periodontal defects in osteoporosis rats, and then the defects were covered with e-PTFE membranes. Defects only covered with e-PTFE membranes were served as control. Histometric measurements with H-E staining were performed after 4 weeks, the formation of new alveolar bone area, cementum area and the width of periodontal ligament were measured and compared. Results Compared with control, the area of newly formed alveolar bone was significantly larger in all experimental groups (P<0.05). The area of new alveolar bone from the transfected BMSCs was significantly higher than that of the non-transfected BMSCs and the control (P<0.05). Transfected cells individually from OVX and SHAM rats showed no significant difference on effects of periodontal regeneration (P>0.05). Conclusion The transfected BMSCs had better effect on repairing the periodontal tissue defects in osteoporosis rats, compared with control.There was no significant difference in bone regeneration between OVX-BMSCs and SHAM-BMSCs.%目的 探讨人骨形成蛋白-7(hBMP-7)基因修饰的大鼠骨髓基质细胞(BMSCs)在骨质疏松(PMO)大鼠牙周组织再生中的作用.方法 通过去除双侧卵巢构建SD大鼠绝经后骨质疏松(PMO)模型,在大鼠右侧下颌第一二磨牙颊侧制备牙周缺损模型.将转染有hBMP-7基因的假手术(SHAM)大鼠的BMSCs和去卵巢(OVX)大鼠的BMSCs,以及未转染的SHAM大鼠的BMSCs和OVX大鼠的BMSCs分别与胶原膜BME-10X构建复合物后植入牙周缺损内.单纯e-PTFE膜组仅覆盖e-PTFE膜.术后4周H-E染色观察牙周组织再生情况.结果 OVX组和SHAM组大鼠的BMSCs均能促进新生牙槽骨的形成,且差异无显著性;经过hBMP-7基因修饰后的BMSCs促进新生牙槽骨修复生成的效果则优于未转染的BMSCs,且在基因修饰后的OVX组和SHAM组BMSCs之间不存在显著性差异;负载BMSCs的组织工程化复合物促进新生牙槽骨修复生成的效果优于单纯胶原膜组.结论 hBMP-7基因修饰后的的BMSCs能更有效地促进PMO大鼠牙周组织缺损的修复,OVX-BMSCs和SHAM -BMSCs具有相似的骨再生能力.
