2例B(A)血型的鉴定

摘要

目的 对2例标本B(A)血型进行鉴定.方法 运用血型血清学、PCR-序列特异性引物(sequence specific primer,SSP)基因定型和ABO基因第6及第7外显子直接测序的方法,对2例标本的B(A)血型和B(A)型等位基因进行检测.结果 2例标本血清学定型分别为A2B和AxB,PCR-SSP法初筛均为B/O基因型,DNA序列分析测定标本1基因型为B(A)02/O02型,标本2基因型为B(A) 02/O01型.结论 采用DNA序列分析法才能准确测定华北地区汉族人群中B(A)血型,PCR-SSP法可能引起差错.%Objective To identify two samples of B(A) blood group. Method To check out B(A) blood group and B(A) alleles of the two samples by serology and PCR-sequence specific primer (SSP) and direct sequencing of ABO gene exon 6 and 7. Result The two samples showed A2B and AxB phenotype serologically and both of them were B/O gene-types by PCR-SSP. DNA sequencing showed B(A)02/O02 and B(A)02/O01 respectively. Conclusion Compared with the error of PCR-SSP , DNA sequencing identifies B(A) blood group accurately in Huabei Han ethnic population.
