针刺对卒中后抑郁大鼠5-HTT、5-HT1AR、NEα2 R蛋白表达的影响

         

摘要

Objective To observe the effect of acupuncture intervention on neurotransmitters and their receptors of post-stroke depression (PSD)rats,so as to explore the mechanism from molecular biological level.Methods Altogether 120 SD male rats randomly divide into blank group (n =10),model group (n =30),acupuncture group (n =40)and medication group (n =40).PSD rat model was induced by middle cerebral artery occlusion(MCAO)and chronic unpredictable mild stress (CUMS).After model-ing,rats of medication group were intragastrically administered fluoxetine at the dose of 2 mg /kg once daily,those of acupuncture group were needled at bilateral acupoints of Baihui (GV 20),Fengfu (GV 16),Shenmen (TF 4),Taichong (LR 3)for 20 min once daily with manipulating needles once.The treatment (medication or acupuncture)were underwent three courses for 21 days .Then protein expres-sion of norepinephrine receptor 2R (NEα2 R),5-hydroxytryptamine receptor1A (5-HT1A R) and 5-hydroxytryptamine transporter(5-HTT)in PSD rat hippocampus,raphe nucleus,locus coeruleus were detected by Western blot.Results Compared with blank group,5-HTT protein expression in hippocam-pus,raphe nucleus and locus coeruleus in model group decreased significantly(P <0.01);NEα2 R pro-tein expression in hippocampus,raphe nucleus,locus coeruleus of model group increased significantly(P<0.01);5-HT1A R protein expression in hippocampus,raphe nucleus of model group increased signifi-cantly(P <0.01),that in locus coeruleus of model group increased(P <0.05).Compared with model group,5-HTT protein expression in hippocampus,raphe nucleus,locus coeruleus of acupuncture group and medication group significantly increased(P <0.01);NEα2 R protein expression in hippocampus,ra-phe nucleus,locus coeruleus of acupuncture group and medication group significantly decreased (P <0.01).5-HT1A R protein expression in hippocampus of acupuncture group and medication group signifi-cantly decreased(P <0.01 ),in raphe nucleus,locus coeruleus of acupuncture group and medication group decreased(P <0.05).Compared with medication group,there were no differences of 5-HTT pro-tein expression,NEα2 R protein expression,5-HT1A R protein expression in hippocampus,raphe nucle-us,locus coeruleus of acupuncture group(P > 0.05).Conclusion Acupuncture can reduce the expres-sion of 5-HT1A R and NEα2 R in post-stroke depression model rats,increase the expression of 5-HTT. This suggests that the effect of acupuncture against depression due to stroke via increasing the content or transportation rate of 5-HT and NE.%目的:观察针刺干预卒中后抑郁(PSD)大鼠神经递质及受体的调节和影响,从分子生物水平,阐述针刺干预卒中后抑郁的分子机制。方法将120只雄性 SD 大鼠随机分为空白组、模型组、药物组、针刺组,其中空白组10只,模型组30只,药物组和针刺组各40只,采用大脑中动脉闭塞和慢性不可预见性温和性应激法复合制备卒中后抑郁模型。药物组予以氟西汀2 mg/kg 溶入生理盐水灌胃,针刺组选取百会、风府、神门、太冲穴20 min,期间行针1次,以上治疗每日1次,共27 d,每7 d 之间休息1 d,采用蛋白免疫印迹(Western blot)法检测卒中后抑郁大鼠海马、中缝核、蓝斑核去甲肾上腺素α2受体(NEα2 R)、5-羟色胺1A 受体(5-HT1A R)和5-羟色胺转运体(5-HTT)蛋白的表达。结果与空白组比较,模型组海马、中缝核、蓝斑核5-HTT 蛋白表达非常显著减少(P <0.01),NEα2 R 蛋白表达非常显著增加(P <0.01);海马、中缝核中5-HT1A R 蛋白表达非常显著增加(P <0.01),蓝斑核5-HT1A R 蛋白表达显著增加(P <0.05);与模型组比较,针刺组与药物组海马、中缝核、蓝斑核5-HTT 蛋白表达显著增加(P <0.01),NEα2 R 蛋白表达非常显著减少(P <0.01);海马中5-HT1A R 蛋白表达非常显著减少(P <0.01),中缝核、蓝斑核5-HT1A R 蛋白表达显著减少(P<0.05)。与药物组比较,针刺组海马、中缝核以及蓝斑核5-HTT 蛋白、5-HT1A R 蛋白、NEα2 R 蛋白表达无显著增加(P >0.05)。结论针刺能降低 PSD 大鼠脑内单胺神经递质5-HT1A R 和 NEα2 R的表达量,提高5-HTT 的含量,进一步提示针刺能调节大鼠脑内单胺类神经递质5-HT 和去甲肾上腺(NE)的含量或转运率。从而发挥针刺抗抑郁作用。
