姜黄素对肝纤维化模型大鼠肝纤维化组织细胞因子的影响

     

摘要

目的 了解姜黄素对实验大鼠肝纤维化组织中的基质蛋白酶-13(MMP-13)和金属蛋白组织抑制因子-1(TIMP-1)的干预.方法 将22只雄性SD大鼠随机分为正常组(n=7)、模型组(n=6)和治疗组(n=9).模型组和治疗组用四氯化碳制备大鼠肝纤维化模型,造模6周治疗组加用姜黄素,在10周末处死各组大鼠,取肝脏进行苏木精-伊红(hematoxylin and eosin,HE)染色,运用半定量逆转录聚合酶链反应(RT-PCR)方法检测MMP-13和TIMP-1的含量.结果 HE染色病理分级明显,治疗组与其他两组比较,HE染色病理分级高于正常组,低于模型组,差异均有统计学意义(P<0.05);而MMP-13mRNA的表达组间变化不大,但TIMP-1mRNA的含量在各组间差异有统计学意义(P<0.05),模型组和治疗组TIMP-1表达水平高于正常组,治疗组的TIMP-1mRNA表达与模型组比较明显下降(P<0.05).结论 姜黄素有抗纤维化作用,其机制可能是通过抑制大鼠肝脏中TIMP-1mRNA表达,减少胶原在肝脏内沉积,从而逆转和阻遏纤维化形成.%Objective To ohserve the effects of curcumin on mRNA expression of matrix metal proteinase-13( MMP-13 ) and the tissue inhibitor of metal proteinase -1( TIMP-1 ) in hepatic: tissues of experimental rats. Methods 22 male SpragueDawley rats were randomly divided into 3 groups : normal group ( n = 7 ), model group ( n = 6 ) and treatment group ( n = 9 ) .Hepatic fibrosis was induced by CCl4 in rats,which were treated with treatment after 6 weeks in the treatment group. All rats were sacrificed by the end of the 10th week , when HE staining of hepatic tissue was performed and the semi-quantitative polymerase chain reaction ( RT-PCR ) was used to assay mRNA expression of MMP-13 and TIMP-1. Results HE staining in the treatment group was significantly different from the model group ( P<0. 05 ); the mRNA expression of MMP-13 among each group was not remarkable different ( P>0. 05 ), but not for the mRNA expression of TIMP-1 ( P<0. 05 ), Comparing to the normal group; TIMP-1 mRNA in the model group and treatment group were obviously elevated ( P<0. 05 ), compared with the model group, while TIMP-1 mRNA of the treatment group was reduced( P<0. 05 ). Conclusion Curcumin reverses liver fibrosis,the mechanism of which associated to reducing the mRNA expression of TIMP-1 , restoring the liver collagen degeneration system gradually.
