HPLC法同时测定益心舒片中丹酚酸B、五味子醇甲和丹参酮ⅡA的含量

         

摘要

Objectives:To establish a method for the quantitative analysis of salvia acid B, schisandrin and tanshinone IIA in Yix-inshu tablets. Methods:HPLC with C18 chromatographic column was used in the determination of salvia acid B, schisandrin and tan-shinone IIA content in Yixinshu tablets, the mobile phase was acetonitrile-0.5% formic acid solution and the detective wavelength was 286 nm, 250 nm and 270 nm respectively for each constituent. Results:The linear range of salvia acid B, schisandrin and tanshi-none IIA were 0.18~2.66μg (r=0.9992), 0.041~0.6140μg (r=0.9999), 0.016~0.237μg (r=0.9998);their average recovery rate (n=6) were 92.2%(RSD=1.9%), 96.9%(RSD=1.7%), 98.6%(RSD=1.3%). Conclusions:The method was simple and accurate. Wa-ter-soluble constituent salvia acid B and lipid-soluble constituent tanshinone IIA were determined in a single analysis, so that the pro-cess of alcohol extraction of Radix salviae miltiorrhizae could be monitored while controlling the manufacturing process of Yixinshu tablets.%目的:建立同一色谱条件同时测定益心舒片中丹酚酸B、五味子醇甲和丹参酮ⅡA的定量分析方法。方法:采用高效液相色谱法。高效液相色谱条件:以十八烷基硅烷键合硅胶为填充剂;流动相:甲醇-乙腈-0.5%甲酸溶液采用梯度洗脱;波长:286nm、250nm、270nm。结果:益心舒片中丹酚酸B、五味子醇甲和丹参酮ⅡA的线性范围依次为0.18~2.66μg (r=0.9992),0.041~0.6140μg(r=0.9999),0.016~0.237μg(r=0.9998);平均回收率(n=6)依次为92.2%(RSD=1.9%),96.9%(RSD=1.7%),98.6%(RSD=1.3%)。结论:本方法操作简便,结果准确,同时检测了水溶性成分丹酚酸B和脂溶性成分丹参酮ⅡA,在控制处方投料的同时还能控制丹参乙醇提取的生产工艺。
