汉黄芩素对人肺癌A549细胞株增殖活性的影响

摘要

Objective:To study the effect of wogonin on proliferation of human lung cancer cells A549 to provide theoretical foun-dation for clinical application. Methods:MTT assay and the cell growth curve were adopted to detect the effects of different concentra-tions of Wogonin on the proliferation activity of lung cancer cells. The morphological changes of A549 cells were observed by HE staining. Results:Wogonin could apparently inhibit human lung cancer cell line. When the drug concentration reached 10μg/mL, the proliferation inhibition was obvious. The cell survival concentration decreased with the time.Wogoin could change the shape of A549 cells, such as cell roundeing, volume reduction, nuclear anachromasis, cytoplasm agglutination.Conclusions:Wogonin could inhibit proliferation of human lung cancer cells A549 and will have a certain guiding significance to the treatment of lung cancer.%目的：本实验通过研究汉黄芩素对人肺癌A549细胞株增殖活性的影响，为其机制的研究及临床应用提供理论依据。方法：实验通过MTT法、生长曲线法、检测不同浓度汉黄芩素、不同作用时间对肺癌细胞增殖活性的影响，通过HE染色实验观察汉黄芩素对A549细胞形态学变化。结果：实验显示汉黄芩素对人肺癌细胞株有较强的抑制作用，当药物浓度达到10μg/mL时，增殖抑制作用明显。随着给药时间延长，细胞存活浓度递减。汉黄芩素能够改变肺癌细胞形态，用药后与对照组比较细胞变圆，体积缩小，核质深染，胞质凝集。结论：汉黄芩素能够抑制人肺癌细胞A549增殖，改变细胞的形态，将对肺癌治疗具有一定的指导意义。
