HPLC法测定清火栀麦滴丸中脱水穿心莲内酯和栀子苷的含量

摘要

Objective To discuss the content of dehydrated andrographolide, geniposide in QingHuoZhiMai dripping pills by high performance liquid chromatography (HPLC). Methods The content of dehydrated androgra-pholide, geniposide was determined by HPLC, with C18 elite (4.6 mm×200 mm, 5 m) as chromatographic column, meth-anol-water (55:45) as the mobile phase and 250 nm as the detection wavelength for dehydrated andrographolide, acetoni-trile-water (13:87) as the mobile phase and 238 nm as the detection wavelength for geniposide. Results Dehydrated an-drographolide in QingHuoZhiMai dripping pill has a good linear relationship in the range of 0.20~1.26μg (r=0.999 9), and the average recovery rate was 98.14%. Geniposide in QingHuoZhiMai dripping pill has a good linear relationship in the range of 1.59~6.46μg (r=0.999 9), and the average recovery rate was 97.36%. Conclusion HPLC presents a fast, simple, and accurate way for detecting the content of dehydrated andrographolide, geniposide in QingHuoZhiMai drip-ping pills, which is worthy of clinical application.%目的：采用高效液相色谱法(HPLC)测定清火栀麦滴丸中脱水穿心莲内酯和栀子苷含量。方法采用高效液相色谱法(HPLC)，以依利特C18(4.6 mm×200 mm，5μm)为色谱柱，以甲醇-水(55：45)为脱水穿心莲内酯的流动相，250 nm为脱水穿心莲内酯的检测波长，以乙腈-水(13：87)为栀子苷的流动相，238 nm为栀子苷的检测波长对清火栀麦滴丸中脱水穿心莲内酯、栀子苷含量进行测定。结果经HPLC法测定，清火栀麦滴丸中的脱水穿心莲内酯在0.20~1.26μg范围区间内具有良好的线性关系(r=0.9999)，平均回收率为98.14%。清火栀麦滴丸中的栀子苷在1.59~6.46μg范围区间内具有良好的线性关系(r=0.9999)，平均回收率为97.36%。结论高效液相色谱法（HPLC）可以快速、简便、准确地测定清火栀麦滴丸中脱水穿心莲内酯、栀子苷的含量，值得应用。
