香菇多糖和黄芪多糖对免疫抑制小鼠免疫功能调节的研究

         

摘要

目的:研究香菇和黄芪多糖对免疫抑制小鼠免疫功能的调节作用.方法:通过检测环磷酰胺免疫抑制模型小鼠的迟发型变态反应程度(DTH,足跖增厚法)、脏器指数、NK细胞活性(乳酸脱氢酶释放法)、淋巴细胞增殖能力(MTT试验)以及IL-2、IL-4、IL-10、IFN-γ水平变化(ELISA试剂盒)来评价香菇和黄芪多糖对免疫抑制小鼠免疫功能的影响.结果:实验表明,环磷酰胺能显著抑制正常小鼠DTH程度、NK细胞活性及细胞因子分泌水平(均为P<0.05);香菇和黄芪两种多糖能显著恢复DTH程度(P<0.05),极显著增高NK细胞包活性(P<0.01),同时能提高血清中IL-2、IL-4、IL-10、IFN-γ的含量.结论:香菇及黄芪多糖对免疫抑制小鼠具有显著的免疫功能恢复效果;香菇多糖的免疫调节效应明显优于黄芪多糖.%Objective: To investigate the effect of Lentinan and Astragalus polysaccharides on immunoregulation in mice . Methods: After oral administration of the two polysaccharides , immune effect of the two polysaccharides on the whole immunity was in -vestigated through delayed type hypersensitivity (DTH) , visceral organs index , NK cell activity (lactate dehydrogenase releasing method) , lymphocyte proliferation capacity (MTT) and the level of IL-2, IL-4, IL-40, IFN-γ( ELISAs) of immunosuppressed mice induced by cyclophosphamide. Results: Cyclophosphamide -treated mice showed a significant decrease in DTH , NK cell activity and the serum level of cytokines (each was P <0. 05) ; Both Lentinan and Astragalus polysaccharides accelerated recovery of DTH (P < 0.05 ) , as well as NK cell activity ( P < 0. 01) and restored the level of IL -2, IL-4, IL40, IFN-γ in the serum. Conclusion: Both Lentinan and Astragalus polysaccharides can significantly stimulate the immune function in mice with immunologic suppression ; the im— munological enhancement effect of Lentinan is obviously better than Astragalus polysaccharides .
