姜黄素对直肠癌CD133+肿瘤干细胞样细胞群放疗敏感性的影响

         

摘要

目的:探讨姜黄素对直肠癌CD133+肿瘤干细胞样细胞群放疗敏感性的影响。方法应用免疫磁珠分选器、阳性分离柱分选出直肠癌CD133+肿瘤干细胞,随机分成姜黄素组、姜黄素联合放疗组、放疗组,应用四甲基偶氮唑蓝比色( MTT)法检测24、48、72 h各组细胞生长抑制情况;膜联蛋白V-异硫氨基荧光素/碘化丙啶( Annexin V-FITC/PI)双染色检测细胞凋亡率。结果姜黄素组随着时间变化肿瘤细胞抑制率变化不大( P>0.05);放疗组和姜黄素联合放疗组24、48、72 h细胞生长抑制率逐渐升高(P<0.05);同期三组细胞生长抑制率均有统计学意义(P<0.05),两两比较24 h细胞生长抑制率,姜黄素组与姜黄素联合放疗组和放疗组之间差异有统计学意义( P<0.05),姜黄素联合放疗组与放疗组之间差异无统计学意义(P>0.05);48、72 h抑制率两两之间差异均有统计学意义(P<0.05);姜黄素组48 h与72 h之间细胞凋亡率差异无统计学意义(P>0.05),姜黄素联合放疗组和放疗组48 h与72 h之间细胞凋亡率之间差异具有统计学意义( P<0.05)。同期内不同组细胞凋亡率之间的差异有统计学意义( P<0.05),两两比较细胞凋亡率之间差异具有统计学意义(P<0.05)。结论姜黄素可以增加直肠癌 CD133+肿瘤干细胞样细胞群放疗敏感性,抑制肿瘤细胞生长促进细胞凋亡。%Objective To investigate the influence of radiotherapy sensitivity of curcumin on CD133+tumor stem cell-like cell popu-lations in rectal carcinoma.Methods The CD133+cancer stem cells in rectal cancer were separated by immunomagnetic beads sorting de-vice and the positive column, and then randomly divided into curcumin, curcumin combined with radiation, radiotherapy groups.The cell growth inhibition were detected in 24 h, 48 h and 72 h by MTT method,and the cell apoptosis rate was detected by Annexin V-FITC/PI double staining.Results The tumor cell inhibition rate of curcumin group was changed little along with the time(P>0.05),the cell growth inhibition rates of radiotherapy group and curcumin combined with radiation group were gradually increased in 24 h, 48 h and 72 h ( P<0.05);the cell growth inhibition rate between three groups was statistically significant over the same period(P<0.05),the difference of cell apoptosis rate was no statistical difference in curcumin group in 48 h and 72 h(P>0.05), while it was significantly different when curcumin combined with radiotherapy group compared with radiotherapy group(P<0.05);the cell apoptosis rate was different in different groups of the same period(P<0.05),and also was significantly different between any two groups(P<0.05).Conclusions Curcumin could increase the radiation sensitivity of CD133+tumor stem cell-like cell populations in rectal carcinoma, restrain tumor cell growth and promote cell apoptosis.
