矽肺观察对象早期蛋白标志物的研究

         

摘要

Objective To study mainly the change analysis of proteomic expression profile between silicosis objects of observation and healthy people,so as to investigate the protein biomarkers of silicosis in early stage.Methods 10 samples fromobservation objects of silicosis and 10 healthy people were collected and performed by two-dimensional electrophoresis to screen protein with differential expression ,and then mass spectrumidentification was analyzed . Results C ompared with non-dust contacThealthy people,39 up-regulated proteins and 7 down-regulated proteins with more than twice of differential expression were successfully identified .In observation objects of silicosis,down-regulated proteins showed transforming growth factorβprecursor,dipeptidyl peptidase 1 ,and apolipoprotein A-Ⅳ precursor,and up-regulated proteins showed interferon βprecursor,tumor necrosis factor,Furin proteinase,deoxyribonuclease Ⅱ, glycoprotein ,granzyme A and chemokine receptor.Conclusion These differently expressed protein factors are mainly involved in immune regulation,cytotoxicity,and metabolism,which plays an important role in the early development of silicosis through immunoreaction and proteinase activity.The results provide some potential protein molecular targets for early clinical diagnosis of silicosis,which are helpful for further study and application of markers of silicosis in early stage.%目的:本研究的目的主要集中在矽肺观察对象与非接尘健康正常人之间蛋白组表达谱的变化分析,探索矽肺发展的早期蛋白标志物。方法收集矽肺观察对象组和健康正常人组样本各10例,利用双向电泳检测筛选差异表达的蛋白,然后进行质谱鉴定分析。结果与非接触粉尘比较,表达差异在两倍以上的39个上调蛋白和7个下调蛋白被成功鉴定出来。在矽肺观察对象组下调的蛋白有转化生长因子β前体、二肽基肽酶1、载脂蛋白A-Ⅳ前体等蛋白;上调的蛋白有干扰素β前体、肿瘤坏死因子、弗林蛋白酶原、脱氧核糖核酸酶Ⅱ、糖蛋白、颗粒酶A、趋化因子受体等蛋白因子。结论这些显著变化的蛋白因子主要参与免疫调控、细胞毒性、物质代谢等过程,可能通过免疫反应和蛋白酶活性在矽肺的早期发展中起重要作用。研究结果为矽肺早期临床诊断提供了一些潜在的蛋白分子靶点,有助于矽肺早期标志物的进一步研究和应用。
