目的 探讨经多聚赖氨酸预处理的花生DNA疫苗对花生蛋白诱导变态反应的预防作用.方法 以编码花生蛋白Arah2的质粒DNA (pArah2)、经多聚赖氨酸(PLL)预处理的质粒DNA( PLL-pArah2)、磷酸盐缓冲液(PBS)和PLL对照皮内注射免疫Balb/c小鼠,1次/周,连续3周,再经腹腔注射纯化的Arah2蛋白免疫小鼠.在不同时间点收集各组小鼠血清,用酶联免疫吸附法检测血清中Arah2特异性IgG1、IgG2a抗体以及血清总IgE抗体水平,分析DNA疫苗免疫对蛋白诱导小鼠变态反应的影响.结果 pArah2及PLL-pArah2质粒DNA免疫后,小鼠血清Arah2特异性IgG1和IgG2a升高,但血清总IgE抗体未见升高.Arah2蛋白免疫后,不同处理组血清总IgE 、Arah2特异性IgG1和IgG2a抗体水平均升高,但pArah2组血清总IgE和Arah2特异性IgG1抗体水平明显低于PBS对照；PLL-pArah2组血清总IgE和Arah2特异性IgG1、IgG2a抗体水平不仅明显低于PBS和PLL对照,且明显低于单纯pArah2组.结论 经多聚赖氨酸预处理的Arah2 DNA免疫能够更好地抑制之后蛋白免疫产生的变态反应应答,为DNA疫苗治疗过敏性疾病提供了新的思路.%Objective To investigate the prophylactic effect of poly-1-lysine modified peanut DNA vaccine on peanut protein induced allergy. Methods The Balb/c mice were immunized at a weekly interval for 3 weeks with intradermal injection of DNA vaccine which encodes peanut protein Arah2 (pArah2) and pArah2 pre-treated by poly-1-lysine ( PLL-pArah2), by using PBS and PLL as controls. Peripheral blood samples were collected from the mice at different time points, and Arah2 specific serum IgGl, IgG2a and total IgE were analyzed by ELISA methods. Results The Arah2-specific IgGl and IgG2a antibodies levels were elevated in the serum of pArah2 and PLL-pArah2 DNA immunized mice, and there were no changes in total IgE level after DNA immunization. However, after arah2 protein immunization, the total IgE and Arah2-specific IgGl antibodies levels in mice pre-immunized with pArah2 were significantly lower than that of PBS group; the total IgE and Arah2-specific IgGl and IgG2a antibodies levels in mice pre-immunized with PLL-pArah2 were significantly lower than that of PBS and PLL groups. The antibodies levels of PLL-pArah2 group were particularly even lower than that of naked pArah2 group. Conclusion The inhibitory effect of DNA vaccine was greatly improved on subsequent peanut protein induced allergy by modifying the peanut DNA vaccine with PLL
