羧甲基壳聚糖水凝胶的合成及其对人皮肤成纤维细胞增殖的影响

         

摘要

A series of carboxymethyl chitosan (CMCS)-genipin hydrogel have been synthesized by adding different concentration of calcium chloride solution.TNBS method was applied to evaluate the effect of the different concentrations of calcium ions on free amino content in the CMCS hydrogel.CCK-8 method was applied to evaluate the effect of the concentrations of free amino in hydrogel on human skin fibroblast proliferation.The results demonstrate that the content of the free amino groups decreases with the increasing concentration of calcium added in the hydrogel, which promotes the proliferation of fibroblasts.Therefore, CMCS-genipin hydrogel may be a potential wound dressings.%制备了一系列羧甲基壳聚糖(carboxymethyl chitosan,CMCS)-京尼平水凝胶.通过向CMCS中加入不同浓度的氯化钙溶液,然后与京尼平进行交联反应合成水凝胶,用三硝基苯磺酸(TNBS)法测定自由氨基含量,考察添加钙离子的浓度对CMCS水凝胶中自由氨基含量的影响.通过在水凝胶上培养人皮肤成纤维细胞,用CCK-8法测定细胞增殖结果,考察了水凝胶中自由氨基浓度对人皮肤成纤维细胞增殖的影响.结果表明,随着添加的钙离子浓度的提高,水凝胶中自由氨基的浓度降低,人成纤维细胞增殖能力提高.
