高效液相色谱法测定犬血浆中磷酸苯丙哌林的质量浓度

摘要

Objective To establish an HPLC method to detect the mass concentration of benproperine phosphate in dog plasma. Methods The mobile phase consisted of 0. 02 mol/L potassium dihydrogen phosphate solution (containing 0.5％ triethylamine, adjusted pH to 4.0 with phosphoric acid)-acetonitrile (60∶40) was used. Nicardipine was used as the internal standard. The plasma samples were pretreated by mixed solvent of N-hexane and avantin, then analyzed by HPLC. Results The linear range was 0.01-0. 80μg/mL( r2 =0. 998). And the average recovery rate was 96. 89％- 109.30％. Both the within-day and between-day RSD were less than 12％. The minimum detective limit was 5 ng/mL. Conclusion This HPLC method is rapid, accurate, and sensitive, which can be applied to the pharmacokinetic study of benproperine phosphate.%目的 建立用于测定犬血桨中磷酸苯丙哌林质量浓度的高效液相色谱法.方法 以0.02 mol/L磷酸二氢钾(含0.5%三乙胺,磷酸调pH至4.0)-乙腈(60:40)为流动相,尼卡地平为内标,血浆样品经正己烷-异丙醉的混合溶剂萃取后,采用高效液相色谱法测定血浆中磷酸苯丙哌林的质量浓度.结果 磷酸苯丙哌林质量浓度在0.01～0.80μg/mL范围内与峰面积比线性关系良好(r =0.998) ;平均相对回收率为96.89%～109.30%,日内和日间精密度RSD均小于12%,最低检测限为5ng/mL.结论 所用方法 快速、准确、灵敏,可用于磷酸苯丙哌林的药代动力学研究.
