丹参多酚酸盐对2型糖尿病大鼠心脏保护作用

摘要

Objective To explore the protection effect of salvianolate in type 2 diabetes rats.Methods Of 30 healthy male Wistar rats,20 were selected and fed high-fat and high-sucrose diet for 6 weeks.They were injected intraperitoneally streptozotocin (STZ,30 mg/kg).After 3 days,the rats with plasma glucose ≥ 17.8 mmol/L were as the rat model of diabetes(n =17) and randomly divided into the diabetes group(n =9) and salvianolate (15 mg/kg)treatment group (n =8).Other 10 heathy male wistar rats were included into the control group (group N,n =10).S group were treated with salvianolate by intraperitoneal injection for 8weeks.N and D groups rats were given saline.Eight weeks later,heart function was detected by Doppler echocardiography ; the pathological changes were observed under transmission electron microscope and light microscopy; the levels of Bcl-2 and Caspase-3 gene expressions were examined by fluorescence quantitative polymerase chain reaction.Results Compared with control group,cardiac myocyte apoptosis was significantly increased (14.57 ± 0.56) vs (0.87 ± 0.13) (P < 0.05) ; Bcl-2 expression was decreased(0.57 ± 0.05) vs (1.06 ± 0.09) (P < 0.05) ; Caspase-3 expression was significantly increased (1.94 ± 0.15) vs (1.02 ± 0.08) (P < 0.05).Compared with diabetes group,after given salvianolate treatment,apoptotic myocardial cells decreased[(6.16 ±0.64) vs (14.57 ±0.56)] (P <0.05) ;the level of myocardial Bcl-2 gene expression increased [(0.84 ± 0.09) vs (0.57 ± 0.05)] (P < 0.05) ; while Caspase-3 decreased [(1.37 ± 0.13) vs (1.94 ± 0.15)] (P < 0.05).Conclusion Salvianolate treatment can delay the development of type 2 diabetic cardiomyopathy and protect the heart.%目的 探讨丹参多酚酸盐对2型糖尿病大鼠心脏保护作用.方法 选择30只健康雄性Wistar大鼠中的20只,以高糖高脂饲料喂养6周后腹腔注射链脲佐菌素30 mg/kg,3d后测血糖≥17.8 mmol/L(n=17)入选糖尿病大鼠模型,并随机分为糖尿病组(n=9)、丹参多酚酸盐(15 mg/kg)治疗组(n=8),另外10只大鼠为对照组(n=10).治疗组给予丹参多酚酸盐治疗8周,彩色多普勒超声检测大鼠心功能,光镜及透射电镜观察心肌病理学改变,转移酶介导的缺口末端标记法检测心肌细胞凋亡,荧光定量聚合酶链反应检测心肌细胞Bcl-2及半胱氨酸天冬酶3(Caspase-3)基因的表达情况.结果 与对照组相比,糖尿病组凋亡心肌细胞明显增多[(14.57 ±0.56)比(0.87±0.13)](P<0.05);心肌中Bcl-2基因表达减弱[(0.57 ±0.05)比(1.06 ±0.09)](P<0.05);Caspase-3基因表达增强[(1.94±0.15)比(1.02±0.08)](P<0.05);与糖尿病组相比,丹参多酚酸治疗组凋亡心肌细胞减少[(6.16±0.64)](P<0.05);心肌Bcl-2基因表达增多[(0.84±0.09)](P<0.05);Caspase-3基因表达减少[(1.37±0.13)](P<0.05).结论 应用丹参多酚酸盐治疗后,可延缓2型糖尿病心肌病变发展,保护心脏.
