大鼠脑出血灶周水肿变化与缺血改变的研究

         

摘要

Objective: To study of the dropsy and ischemia charging around the hemorrhage regon, the changing of brain water content, ischemia region,apoptosis on rats cerebral hemorrhage models are observed in different time spot. Methods:After the body artery blood being injected into the rats' brain, testing the brain water content; dyeing with TTC; examining the Apoptosis with TUNEL in different time spot, 3 d, 5 d, 7 d, 14 d, 21 d. To compare with that the change of the dropsy and ischemia around cerebral hemorrhage in different time spot. Results: TTC dyeing, around the hematoma the color of infarction space expands gradually 14 d later was remarkahle [3 d group [(0.31±0.06)mm; 14 d group (2.03±0.13)mm] (P<0.01). The hematoma area reduces gradually. 14d later was remarkable [3 d group (19.17±1.52) mm2;14 d group (6.91±0.59) mm2] (P<0.01). Brain water content, the experiment group was higher than the control group [3 d experiment group (82.23±2.71)%; 3 d control group (68.56±1.70)%] (P<0.01); 3-7 d the brain water content arrives at the maximum level, then it droped and it was roughly invariable after 14 days 13 d group (82.23±2.71)%; 14 d group (72.98±1.81)%] (P<0.01). Apoptosis, the experiment group was higher than the control group [3 d experiment group (20.25±2.50);3 d control group (1.90±0.51)] (P<O.01); 3-14 d apoptosis continue to express high. 14 days later started to drop [3 d group (20.25±2.50); 14 d group (15.55±2.60)] (P<O.01). Conclusion: The region around cerebral hemorrhage in rats is one kind of dropsy and ischemia mixing zone. 3-14 days it is mainly the dropsy, after 14 days, dropsy is roughly constant, it is mainly the ischemia.%目的:通过观察大鼠脑出血模型不同时段的脑含水量,灶周缺血区,凋亡细胞指标的改变,探讨血肿灶周水肿、缺血的变化.方法:自体动脉血注入法制作大鼠脑出血模型,于脑出血(ICH)损伤后3、5、7、14、21 d时间点进行脑含水量测定,2,3,5-氯化三苯基四氮唑(TTC)染色,原位末端标记(TUNEL)法检测凋亡细胞,应用图像处理软件,结合病理组织学改变,比较各个时间点ICH模型血肿灶周水肿与缺血的演变过程.结果:TTC染色,血肿周围色淡染,梗死带内径逐渐扩大,14 d后变化最为显著[3 d组(0.31±0.06)mm;14 d组(2.03±0.13)mml(P<0.01).手术组3~21 d血肿面积逐渐减小,14 d后变化显著[3 d组(19.17±1.52)mm2;14 d组(6.91±0.59)mm2](P<0.01).脑含水量,手术组含水量均高于对照组13 d手术组(82.23±2.71)%;3 d对照组(68.56±1.70)%1(P<0.01).手术组3~7 d脑含水量达最高值,而后下降,至14 d后基本恒定[3 d组(82.23±2.71)%;14 d组(72.98±1.81)%1(P<0.01).凋亡细胞,手术组均高于对照组[3 d手术组(20.25±2.50)个;3 d对照组(1.90±0.51)个](P<0.01);手术组3~14 d凋亡细胞持续高表达,14 d后开始下降[3 d组(20.25±2.50)个;14 d组(15.55±2.60)个](P<0.01).结论:大鼠脑出血后,灶周是一种水肿缺血混合区域,3~14 d以水肿为主,14 d后水肿相对恒定,以缺血为主.
