抗宫炎凝胶剂的提取工艺研究

         

摘要

目的:优选七类新药抗宫炎凝胶剂的提取工艺。方法采用紫外可见分光光度法分别测定广东紫珠中的总黄酮、益母草中的盐酸水苏碱含量;采用高效液相色谱法对乌药中乌药醚内酯的含量进行测定。分别以总黄酮、盐酸水苏碱、乌药醚内酯的含量及干膏得率为考察指标,对处方中三味药进行提取工艺优化。利用正交试验设计法,以提取次数、提取时间、溶媒量、乙醇浓度为4个因素,每个因素设计3个水平。结果广东紫珠的最佳提取工艺为10倍量50%乙醇回流提取3次,每次3 h;益母草的最佳提取工艺为10倍量95%乙醇回流提取2次,每次1.5 h;乌药的最佳提取工艺为8倍量85%乙醇回流提取2次,每次1 h。结论优选的提取工艺科学合理,适于提取工艺中多因素的优选,可作为新药抗宫炎凝胶剂的工业化生产提取工艺,为制备安全稳定、速释高效新剂型的研究奠定科学基础。%Objective To optimize the extraction process of the seventh drug for Kanggongyan Gels. Methods UV-visi-ble spectrophotometry was used to determine the contents of total flavonoids in Guangdongzizhu and stachydrine hy-drochloride in Yimucao; HPLC was adopted to determine the contents of lindera ether in Wuyao. The contents of total flavonoids, stachydrine hydrochloride, lindera ether ester as well were taken as extract rate as assessment index to opti-mize the extraction process of the shamisen drugs in prescription. By using orthogonal experimental design method, the extraction times, extraction time, solvent usage and the concentration of ethanol as factors were taken, and three levels of each factor were designed. Results The best program for extraction of Guangdongzizhu was 10 times of 50% ethanol extraction in 3 h each for 3 times; the best program for extraction of Yimucao was 10 times of 95% ethanol extraction in 1.5 h each for 2 times; the best program for extraction of Wuyao was 8 times of 85% ethanol extraction in 1 h each for 2 times. Conclusion The optimal extraction technology is reasonable and scientific, and it can be used for optimiz-ing factors in extraction technology and industrial production. For researching the preparation of security and stability, immediate-release dosage form of efficient lays the scientific foundation.
