为了研究姜黄和郁金的寒热药性差异,采用HPLC-MS建立了大鼠脑中7种儿茶酚胺类物质的分析方法,对寒、热证候模型及采用姜黄与郁金干预后相应模型中大鼠脑中儿茶酚胺类代谢物进行了分析.实验数据采用主成分分析(PCA)进行处理,采用方差分析(ANOVA)判别主要差异代谢产物.结果表明,寒、热模型和正常对照组通过主成分得分图(PCA Scores Plot)可分为3个部分,主要差异代谢物为肾上腺素(E),3-甲氧基酪胺(3-MT)和5-羟色胺(5-HT),说明大鼠脑中儿茶酚胺类成分与寒、热因素存在相关性; 姜黄对寒证模型的作用较为明显,主要差异代谢物为肾上腺素(E)和3-MT,说明姜黄具有温性药性的性质; 郁金和姜黄对热证模型均有一定作用,但郁金的作用强于姜黄,主要差异代谢物为肾上腺素(E)、3-MT、5-HT和3,4-二羟基苯乙酸(DOPAC),说明郁金具有寒性药性的性质.结果表明,单胺类神经递质代谢谱的变化可用于评价姜黄和郁金寒热药性的差异.%To compare and confirm different effects of Jianghuang and Yujin, a HPLC-MS method for the quantitative determination of 7 catecholamines in brain samples was developed. Principal component analysis (PCA) was processed to analyze the catecholamine metabolic profiling. The results suggested that heat-syn-drome model and cold-syndrome model could be distingushed by catecholamine metabolic profiling. Jiang huang and Yujin have different effects for the heat and cold syndrome model based on brain catecholamines metabolic profiling.
