不同培养基质和培养时间对蛹虫草子实体生物量和蛋白质氨基酸组成的影响

         

摘要

研究了不同培养基质和不同培养时间对蛹虫草栽培菌株CM-H0810子实体生物量和氨基酸含量、组成的影响,以期为蛹虫草的生产及充分开发利用提供参考.结果表明:使用培养基E(蚕蛹)栽培获得的子实体生物量最低,但氨基酸总量最高,且必需氨基酸和非必需氨基酸比值以及必需氨基酸和氨基酸总量比值较为理想;与培养基C(米饭)比较,培养基A(麦粒)栽培获得的子实体生物量、蛋白质含量、氨基酸总量均较高;在培养基A(麦粒)或C(米饭)中添加10%蚕蛹粉后,子实体生物量变化不明显,但蛋白质含量和氨基酸总量显著高于未添加蚕蛹粉的处理;培养基A(麦粒)接种后第55天子实体生物量最高,第60天氨基酸总量最高;各供试培养基栽培的蛹虫草子实体,虽均以Leu为限制氨基酸,但氨基酸比值系数排序有所不同;不同培养时间的子实体样品中,氨基酸比值系数排序均为Thr>Phe+Tyr>Lys>Val>Ile>Leu>Met+Cys.%In order to provide reference for the full development and utilization of Cordyceps militaris,the effects of different culture media and culture time on the biomass and amino acid content and composition of fruiting body of C. militaris cultivated strain CM-H0810 were studied. The results showed that the biomass of fruiting body cultured by culture medium E (silkworm chrysalis)was the lowest,but the total amino acid content was the highest,and the ratio of essential amino acids to nonessential amino acids and the ratio of essential amino acids to total amino acids were ideal.Compared with culture medium C (rice),the biomass,protein content and total amino acids of fruiting body cultured by culture medium A (wheat kernel)were higher.After adding 10% silkworm chrysalis powder to culture medium A (wheat grain)or C (rice),the change of fruiting body biomass was not obvious,but the protein content and total amino acids were significantly higher than those of non silkworm chrysalis powder treatment.The highest biomass of fruiting body was obtained in the fifty-fifth day cultured by culture medium A (wheat kernel),and the highest amino acid content was obtained in the sixtieth day.Although Leu was the limiting amino acid of all the fruiting bodies of C.militaris cultured by different culture media,but the order of the amino acid coefficient was different. The amino acid coefficients of fruiting body samples with different growth time were all Thr>Phe+Tyr>Lys>Val>Ile>Leu>Met+Cys.
