Recent success in the induction of pluripotent stem cells from adult somatic cells provided a promising tool for regenerative medicine and tissue engineering.Human induced pluripotent stem cells(hiPSCs)exhibit similar characteristics to embryonic stem cells,including the ability to differentiate into cell types of three germ layers(1).Despite various successful applications,many challenges in hiPSCs manufacturing processes need to be addressed.One challenge is the difficulty in preparing a uniform cell population in high-density seeding of adherent hiPSCs,leading to cell-cell variation of cell products.Currently,there is a limited understanding of cell culture in high-density seeding,where this method is applied in scaling up and preparation of favorable cell environment for differentiation into some cell lineages(2).In the current study,we proposed a method to quantify spatial uniformity of cell population and compare different culture conditions for high-density seeding process.Ultimately,we aim to understand the effect of spatial uniformity on subsequent stages of cell production.
