摘要

The purpose of this paper is to observe the influence in rats with Parkinson's disease PD treated by Neural stem cells (NSCs) modified by pIRESneo-EGFP-GDNF-TH. pIRESneo-EGFP-GDNF-TH plasmid was established by reconstructing pEGFP(Nl)-GDNF plasmid and pIRESneo, NSCs was transfected by electroproation. PD rat models were set up by 6-OHDA and then divided into six groups randomly: sham group, PD group, NSCs group, GDNF group, TH group, and GDNF-TH group. Rotating behaviors of rats were induced by intraperitoneal injection of apomorphine at 2, 4, 8 weeks after NSCs transplantation or NSCs modification of three kinds of plasmids through cerebral lateral ventricle. Transplantation of NSCs modified by pIRESneo-EGFP-GDNF-TH through cerebral lateral ventricle can increase GDNF and TH protein levels and improve PD rats' behavior.
