The cyclic physiological and anatomical changes in the oviduct is mediated by the local countercurrent transfer of the ovarian products. Thus, cow oviductal epithelial cells (COEC) culture were utilized to investigate the effect of ovarian products such as progesterone (P4), estradiol 17#beta# (E2) and oxytocin (OT) on the oviductal prostaglandin E2 (PGE2), F_2#alpha# (PGF_2#alpha#) and endothelin -1 (ET-1) production. COEC were collected from non-pregnant Holstein cows during the follicular phase and cultured in M199 under standard culture conditions until monolayer formation. Cells in the first passage were incubated with P_4 (500 ng/ml), E_2 (1 ng/ml), OT (10-~9 M) and combination of E_2+P_4 for 24 and 48 h. Administration of E_2 significantly increased the production of PGE_2, PGF_2#alpha# and ET-1. However, simultaneous administration of high level of P_4 blocked the effect of E_2. OT showed no effect on oviductal production of neither PG nor ET-1. These results may imply that locally transferred E_2 at high concentration from the developing follicle is important stimulus for the oviductal PGE_2, PGF_2#alpha# and ET-1 production, which in turn may regulate the oviductal contraction to ensure an optimal embryo transport during the peri-ovulatory period.
