Rat heme oxygenase-2 (HO-2) contains two Cys-Pro pairs, called heme regulatory motifs (HRMs), that regulate the enzyme activity by interacting with heme, but the mechanism is still unknown. We synthesized a 30-residues peptide containing two HRMs of rat HO-2 and its analogs. Electron spin resonance analysis suggested that the thiol group of Cys in HRM is directly coordinated to the ferric heme iron.
