A HR LC-MS method was developed for the comprehensive separation, identification and relative quantitation of lipid classes in biological matrices. The profiling method was able to identify lipids from the 6 LIPID MAPS classes found in serum. All ion fragmentation studies allowed for the characterization of individual lipid side chains using exact mass database searches and chromatographic alignment of fragments (Figure 5). A total of 86 unique TG molecules, representing 81 unique masses, were detected and quantified in the rat sera samples. (Figure 6). 71 TG molecules were able to be characterized using all ion fragmentation (Figure 6). These identifications are significant advances in that they: (i) show greater TG identifications using a non-targeted technique than previously described targeted TG analyses. (ii) characterize the carbon-to-double bond ratio of the 3 TG acyl side chains giving a more detailed insight into the diet-associated modulation of individual TG molecules. A correlation between TG (16:0/16:0/18:1) and TG (16:0/18:1/18:1) and dietary fat composition was established (Figure 7).
