Co-extracted phospholipids from rat plasma samples following sample preparation using either PPT filter or SPE plate may result in severe ion-suppression. The use of a Hybrid SPE-PPT plate provided higher selectivity towards phospholipids under the optimized sample preparation conditions presented. Using the Hybrid SPE-PPT plate and optimized conditions, a revamped LC-MS/MS method was validated that demonstrated improved method ruggedness and provided twice the sample throughput compared to the pre-existing method.
