A SPE of N-linked glycopeptides technique was adopted to extract the N-glycosites from two subsets of PTC and OICR serum samples. Candidate biomarkers were analyzed by HLPC-nanoESI-MS/MS operated in SRM mode. Clustering analyses is providing initial insights into what appears to be a panel of diagnostic biomarkers for PTC samples. OICR analyses is currently ongoing. This analytical approach provides semi-quantitative proteomics results, however, the following issues are currently being addressed in order to improve the quality of results: (1) Inter batch C.V.percent seem to be excessively high (average >48percent). (2) High non-specific binding of tryptic non-glycopeptides (eg. Serum albumin).
