Peptide Hormone Acquisition through Smart Sampling Technique-Mass Spectrometry (PHASST-MS) is a platform designed to identify bioactive peptides. In the PHASST-MS approach, in vitro cell culture models are enriched in endocrine cells. These "smart samples" are used to produce conditioned media containing peptide hormones secreted in response to physiological and pharmacological stimulants. The approach involves no cellular lysis and consequently produces less intracellular protein background. Secreted products up to 16 kDa are directly analyzed and identified during high resolution LC/MS. Through the use of mass maps for label-free relative quantification, features reflective of cellular state or disease state are flagged (Figure 1). Herein, we report preliminary observations on human pancreatic islet cell cultures derived from normal and type 2 diabetic individuals.
