Twelve highly abundant proteins of HFF were successfully captured by IgY-12 columns. OHSS+ and OHSS- samples of HFF (18 each) were analysed in parallel using LC-MALDI-TOF MS and LC-ESI-FTICR MS. LC-MALDI-TOF MS served as a tool for building up the database of major proteins of depleted HFF. DTB contained about 250 protein entries and about 120 unique proteins were typically identified in single LC-MALDI-TOF MS analysis. LC-MALDI data evaluation was focused on significant differences between OHSS+ and OHSS- HFF. The comparison of identified protein lists revealed 5 candidate proteins for quantitation using LC-ESI-FTICR MS. Based on extracted ion chromatograms four of these proteins were found as up-regulated in HFF of patients diagnosed with the syndrome. The candidate proteins will be verified by immunoblotting and AQUA strategy.
