Protein subtraction can drastically improve the absolute number and dynamic range of spectral counts for lesser abundant proteins. Hydrophobic tagging in plasma decreases competition by suppressing the ionization of cysteinyl peptides and increasing LC peak capacity. Protein subtraction and ALiPHAT reagents significantly increase the detection of BNP tryptic peptides in and the number of observed spectral counts in plasma.
