Developed an LC-MS method capable of measuring 2 adducts in 108 nucleosides using only 1.25 (mu)g of DNA. Successfully applied the methodology to quantify adduct levels in 4-ABP dosed RT-4 human bladder cells, observing adduct counts ranging from 10 adducts in 108 nucleosides to 22 adducts in 105 nucleosides. A dosing study involving DNA extracted from rat bladders yielded dG-ABP adduct levels starting at 80 adducts in 108 nucleosides. Identical dose concentrations produced similar adduct levels in both cells and rats. A positive correlation between dose and DNA adduct detection was observed in both cells and rats. dA-ABP was detected in 4-ABP dosed RT-4 cells.
