A simple analytical method based on the MDMS-SL strategy was established to identify and quantify FAHFA species present in biological samples by multiple NLS of the branched FA moiety. This method possessed nearly 4 orders of linear dynamic range, and a limit of detection as lower as 345 amol/μL. Through determination of the FAHFA species levels in liver tissues between db/db mice and their WT controls, we found that the reduced FAHFA amounts in db/db mice may contribute to their insulin-resistance and lead to diabetes.
