Heavy stable isotope labeled internal standard proteins are advantageous for absolute quantitation. Targeted selected reaction monitoring and identification of proteins can be performed in a single assay, shown here with plasma RBP4, VitD-BP, ApoAI and TTR. argeted quantitation data strongly correlated with relative abundance data (iTRAQ). Plasma RBP4 and TTR correlated with vitamin A and Apo-AI correlated with plasma levels of vitamin E, as predicted from relative abundance analysis. Vitamin D and its quantitated binding protein did not, a difference that is currently under study.
