A quantitative procedure for the determination of posaconazole in human fingernails or toenails has been successfully validated for use at Covance. The method utilizes a sample size of 10 mg over the concentration range of 2.5 to 50 ng/10 mg of sample, including a tenfold dilution into this range using blank matrix. The procedure requires complete liquefaction of the nails prior to extraction and allows for the storage of excess liquefied nail solution for potential future assay. To date, this method has been successfully used to analyze >700 samples in support of clinical studies.
