During early stages of therapeutic monoclonal antibody development, titer and cell growth are used to select candidate cell lines. To ensure the selected cell lines are producing protein with the correct amino acid sequence, high sensitivity tryptic mapping techniques are utilized in conjunction with electrospray MS and MS/MS detection to verify the sequence. Occasionally, amino acid substitutions can occur in the protein sequence and are observed experimentally at the peptide level. In this poster, we will present four different software based tools that can be used to search for sequence variants at the peptide level from the extensive data generated during these MS and MS/MS experiments.
