This research is focused on the isolation of D1-protein for further investigation of interaction with phytotoxic compounds and development of new biosensor concepts. This membrane bound protein of the photosystem II (PSII) of the electron transport system in higher plants has been purified and concentrated for the detection of phyto-toxic ligands. D1 is the natural binding partner from numerous phytotoxic compounds such as PSII herbicides and can therefore be applied for their biosensor ic detection.It was of interest to compare the protein composition of the photo synthetic apparatus from different plants in order to find the species with the best properties for the biosensoric application. Therefore thylakoid membranes from four plant species and two different thylakoid-isolation methods have been characterized using green-native 2-D-gel-electrophoresis (green native/SDS-Page).
