Previous attempts at accelerating in vivo bone formation from implantation of mouse adipose derived stromal cells involved exposing these cells, seeded on an apatite-coated PLGA scaffold, to four continuous weeks of retinoic acid exposure. Implantation of these cells into critical-sized calvarial defects resulted in rapid healing followed by osteoclast-mediated bone resorption. This was thought to be mediated by residual retinoic acid on the scaffolds. As a follow-up study, exposure of mouse ASCs in monolayer to shorter courses of retinoic acid, interestingly, revealed accelerated os-teogenic differentiation in vitro. This study seeks to investigate whether such an approach can be translated into the in vivo setting.
