Allergic respiratory disease in adults is associated wiT_H active T-cell immune responses to inhaled allergens that is skewed towards the T_H2 phenotype in contrast to a T_H1- skewed immunity in normal healthy subjects. T_H1 cells enhance cellular immune responses, whereas TH2 cells favor humoral antibody production. Dendritic cell subsets promote either T_H1 or T_H2 via either direct inhibitory effect of IL-10 on T_H1 differentiation or indirectly via IL-10-induced regulatory T cells. The transcription factors T-bet (T-box expressed in T cells) and GATA-3 regulate the function of polarized Thl and Th2 cells, respectively. Newer therapies in allergy and asthma are focused on the development of safer and better immunomodulators to regulate T_H1/T_H2 polarized immunity.
