More reactive oxygen species (ROS) are produced by tumor cells than by normal cells (Szatrowski and Nathan., 1991). Our previous studies have shown that ROS potentiate the invasive activity of rat ascites hepatoma cell line of AH109A, and some food factors which have antioxidative property cancel the ROS-induced invasion (Kozuki et al., 2000, 2001a,b; Zhang et al., 2000). Resveratrol (trans-3,5,4'-trihydroxystilbene), is found in grapes and other foods, and is a polyphenol possessing a strong antioxidative capacity (Jang et al., 1997). We reported that, although resveratrol suppressed both the proliferation and invasion of hepatoma cells at high concentrations, a lower dose (50 μM) of resveratrol or resveratrol-loaded rat sera inhibited only the invasion (Kozuki et al., 2001b). We also found that hepatocyte growth factor (HGF) secretion was enhanced by ROS treatment in AH109A cells (Miura, Y. et al., 2003). HGF has been identified as a motility factor and is secreted by invading AH109A cells. To clarify the mechanisms for the suppressive action of resveratrol and its metabolite(s) on hepatoma invasion, we estimated the intracellular peroxide levels and HGF secretion in AH109A cells.
