摘要

The accumulated antibody concentration of the Fed-Batch culture in Fermenter of the NSO-GS host cell decreased to half the level (300 μg/mL) of the final antibody concentration for flask scale cultivations. Through the various evaluations of characteristic parameters in the Fermenter culture, we found out that it is highly effective in optimizing agitation rate to improve the total cell numbers and keeping low dissolved CO_2 concentration to recover the productivity. Moreover, the cell number was raised by modified medium, which achieved three times greater than original medium. By improving these points in Fermenter culture, finally, accumulated antibody concentration reached 1.5 g/L, an increase of 5-fold compared with the previous method. In this paper, I would like to introduce the data and improvements on the culture system during the establishment of manufacturing scale fermentation system.
