The third-generation chromosome image analyzing system (CHIAS III) was developed using an ordinary personal computer with public domain imaging software, NIH Image, Uneven condensation patterns (CP) appearing on small chromosomes at the mitotic prometaphase stage were analyzed. A quantitative chromosome map or idiogram was developed by the CHIAS III. The analysis consists of three major steps as follows: ①Manipulation of original chromosome images and measurement CP, ②calculation of an averaged CP of homologous chromosomes, and ③development an idiogram based on the averaged CP. The CPs had been analyzed quantitatively by using the first and the second-generation CHIAS with the hard and software specific to the systems . The data obtained using CHIAS III are consistent with those obtained with the chromosome image analyzing systems previously developed. CHIAS III is opened to the public on the Internet ( http://133.1.131.81/Eudejas/chias3/Chias3. html). Quantitative chromosome maps based on the CP Using CHIAS III have been developed in several plant species, for example Oryza sativa, Arabidopsis thaliana, Saccharum spontaneum, Spinacia oleracea, etc. Other applications have also been developed through the CHIAS analysis of FISH signals, idiogram viewer by JAVA, pachytene version. It is anticipated that the development of CHIAS and stepwise new applications to the public will greatly contribute for quantitative analysis of chromosome information.
