The intensive karyotype analysis for a Korean cucumber cultivar (Cucumis sativus L. cv. Winter Long) has been carried out with different methods, such as Feulgen staining, Giemsa C-banding and fluorescence in situ hybridization (FISH). The mitotic chromosomes of the cucumber (2n = 2x = 14) were characterized on the basis of length and arm ratio value. C-banding analysis showed dark stains on the centromeric, telomeric and intercalary regions of chromosomes , except chromosome 2 with a heavy staining in the long arm region. Bicolor FISH using 45S and 5S rDNA probes provides additional information to identify the cucumber chromosomes. The signals for 45S rDNA were detected on the pericentrimeric regions of chromosome 1, 2 and 4, while the signals for 5S rDNA on the short arm of chromosome 5. The similar band patterns as C-banding were observed when chromosomes were counter-stained with 4', 6-diamidino-2-pheny-oindole (DAPI) . All data imply that the karyotype of the Korean cucumber cultivars seems to be peculiar and different from previous reports.
