Using the modular approach of nanoparticle synthesis we had previously published, we successfully created CD133 targeting, CY5 fluorescent, Au@SiO_2 core-shell NPs with >12h of persistence in the blood pool of mice. The nanoprobes successfully targeted CD133(+) ovarian cancer cells in vitro and more importantly, the in vivo tumor location and margin could be determined noninvasively by CT and was correlated with in vivo fluorescence imaging. We were also able to quantify the mass of nanoparticles per unit mass of tumor tissue therefore allowing us to estimate the targeting efficiency of our probes. The data obtained from this study can be used to further optimize targeting strategies to not only image disease sites but also deliver therapeutic molecules for the broad detection and treatment of multiple diseases.
