We have proposed a new algorithm for gel-based DNA computer, which uses the actual biological DNA segments cut by restriction enzymes. Biological DNA segments longer than 1000 base pairs makes it possible to encode huge information over petabytes, although the existing methods such as that of Adleman with artificial short DNA could not. Thus, this algorithm first enables to solve complex problems, An accurate control strategy of polyrnerase chain reaction (PCR) is proposed as the key step for realizing the present algorithm of DNA computing.
