Objective:To study the effect of Shenggu Migu Fang on lumbar bone mineral density(BMD),type I collagen N end propeptide(PINP),alkaline phosphatase(AKP),Osteogenic crosslinking and osteocalcin in serum in ovariectomized rats(OVX). Methods:bilateral ovaries of 60 healthy and clean female SD rats of 6 months old were taken out,and the animal models of osteoporosis were established.Then they were randomly divided into 3 groups,20 in each group:group OVX, group OVX+TCM,group OVX+E2.Drug intervention was carried out at 35 d postoperatively.After 12 weeks,the following changes were measured:BMD measured by bone densitometer,and the serum markers such as AKP,BGP,osteogenic crosslink and PINP detected,and the results were com-pared between the 3 groups.Results:After treatment,there was no significant difference in lumbar BMD measurement between group OVX+TCM and group OVX+E2(P >0.05).In group OVX,the BMD was significantly lower than that of the other 2 groups(P0.05).The BGP level of group OVX+E2and OVX+TCM group was significantly higher than that before operation, while the serum osteogenic crosslinking,PINP and AKP levels were significantly lower than that before the operation(P 0.05).Conclusion:The effect of treating osteoporosis(OP)is remarkable,and it can increase BMD and promote bone formation by improving the biochemical indexes of bone metabolism.%目的:研究生骨密骨方对卵巢去势(OVX)骨质疏松大鼠腰椎骨矿物质密度、I 型胶原N端前肽(PINP)、碱性磷酸酶(AKP)、血清骨原交联及骨钙素(BGP)水平的影响.方法:取6月龄健康清洁级雌性SD大鼠60只,手术摘取雌性大鼠双侧卵巢,建立骨质疏松大鼠动物模型.建模后按随机数字表法分为3组各20只:OVX组、OVX给予生骨密骨方(OVX+TCM)组、OVX给予雌二醇(OVX+E 2)组.术后35 d起分别进行药物干预,12周后测定比较各组以下变化:利用骨密度仪测量腰椎骨密度(BMD),检测血清标志物如AKP、BGP、骨原交联、PINP,比较3组之间结果.结果:治疗后,OVX+TCM组与OVX+E2组腰椎BMD测量差异无显著性(P >0.05),而OVX组腰椎BMD均明显低于其它2组(P 0.05),手术后OVX+E2组及OVX+TCM组BGP水平均显著高于手术前,而血清骨原交联、PINP、及AKP水平均显著低于手术前(P 0.05).结论:生骨密骨方治疗骨质疏松症(OP)效果显著,且生骨密骨方可通过改善骨代谢生化指标,增加BMD,促进骨形成.
