    • 摘要: Neural crest-derived mesenchymal stem cells(MSCs)are known to play an essential function during tooth and skeletal development.PRX1+cells constitute an important MSC subtype that is implicated in osteogenesis.However,their potential function in tooth development and regeneration remains elusive.In the present study,we first assessed the cell fate of PRX1+cells during molar development and periodontal ligament(PDL)formation in mice.Furthermore,single-cell RNA sequencing analysis was performed to study the distribution of PRX1+cells in PDL cells.The behavior of PRX1+cells during PDL reconstruction was investigated using an allogeneic transplanted tooth model.Although PRX1+cells are spatial specific and can differentiate into almost all types of mesenchymal cells in first molars,their distribution in third molars is highly limited.The PDL formation is associated with a high number of PRX1+cells;during transplanted teeth PDL reconstruction,PRX1+cells from the recipient alveolar bone participate in angiogenesis as pericytes.Overall,PRX1+cells are a key subtype of dental MSCs involved in the formation of mouse molar and PDL and participate in angiogenesis as pericytes during PDL reconstruction after tooth transplantation.

    • 摘要: Background:Diffuse large B-cell lymphoma(DLBCL)is typically treated with rituximab,cyclophosphamide,doxorubicin,vincristine,and prednisone(R-CHOP).However,only 60%of patients are cured with R-CHOP.Polatuzumab vedotin is an antibody-drug conjugate targeting CD79b,which is ubiquitously expressed on the surface of malignant B cells.Methods:We conducted a double-blind,placebo-controlled,international phase 3 trial to evaluate a modified regimen of RCHOP(pola-R-CHP),in which vincristine was replaced with polatuzumab vedotin,as compared with standard R-CHOP,in patients with previously untreated intermediate-risk or high-risk DLBCL.Patients 18 to 80 years of age were randomly assigned in a 1:1 ratio to receive six cycles of either pola-R-CHP or R-CHOP,plus two cycles of rituximab alone.The primary end point was investigator-assessed progression-free survival.Secondary end points included overall survival and safety.

    • 摘要: Ionizing radiation can induce endoplasmic reticulum(ER)stress in tumor cells under certain circumstances.How the ER stress affects the radiosensitivity of tumor cells with different p53 status,and ultimately determines the therapeutic effect against cancer,remain poorly understood.Three hepatocellular carcinoma(HCC,Hep G2,Hep 3B and PLC/PRF/5)cell lines were used in this study.Firstly,the radiosensitivity of HCC with different p53 status were assessed by cell proliferation and survival curve.Secondly,whether radiation induces ER stress was analyzed with microscopy imaging and flow cytometry.Finally,the molecular mechanism of how ER stress regulates autophagy,apoptosis and ferroptosis will was studied by microscopy imaging,cellular and molecular biological methods.We aim to elucidate the mechanism of PERK signaling pathway of radiosensitivity of HCC.

    • 摘要: Objective: To evaluate whether the inoculation of contaminated cultures in the peritoneal cavity of mice could implement decontamination of Acanthamoeba cultures.Methods: Suspensions of Acanthamoeba, Acanthamoeba polyphaga ATCC 30461, or Acanthamoeba spp. isolated from soil(Un B13 strain) were inoculated in the peritoneal cavity of Swiss mice(n = 24). After 1, 6, 12, or 24 h of exposure the peritoneal cavity was washed and assessed for the presence of bacteria, fungi, and Acanthamoeba.Results: After 1 h of intraperitoneal inoculation at least 97% of the bacteria and 96% of the fungi(P < 0.05) and 99% of the bacteria(P < 0.05) were successfully eliminated from the ATCC 30461 strain and from the soil isolate Un B13 strain, respectively. This method also allowed the recovery of most trophozoites and cysts from both Acanthamoeba cultures at the end of 24 h.Conclusions: Our data demonstrated that this technique has great potential for decontamination of Acanthamoeba cultures in a short period of time.

    • 摘要: Objective: To evaluate the effect of ethanolic extract of chamomile on balance and motor learning in rats receiving scopolamine and intact rats.Methods: Fourty-two rats were divided into 6 groups(n = 7). Control group received distilled water. Rats in Group 2 were given 1 mg/kg scopolamine. Groups 3 and 4received chamomile extract 200 mg/kg and 500 mg/kg, respectively, and scopolamine simultaneously for 20 days. Intact groups(Groups 5 and 6) only received chamomile extract 200 mg/kg and 500 mg/kg, respectively. Motor coordination of rats was assessed with rotarod apparatus.Results: According to the obtained results, compared with the control group, scopolamine significantly decreased time spent on rotarod performance(P < 0.001). Compared with scopolamine group, the strength and staying on rotarod apparatus in Group 3significantly increased(P < 0.05). The results of this research showed that intact groups that received only chamomile extract at doses of 200 mg/kg and 500 mg/kg significantly increased time spent on rotarod, compared with scopolamine group(P < 0.001).Conclusions: The results of this study indicated the high antioxidant property and protective effect of chamomile extract on motor coordination in the groups that received scopolamine.

    • 摘要: AIM:To investigate the correlation of facet joint osteoarthritis(FJOA) at lumbar levels L4/L5 and L5/S1 and the Oswestry Disability Index(ODI).METHODS:The study involved lumbar MRIs of 591 patients with a mean age of 47.3 years.The MRIs of the lumbar spine were performed on a 1.5 Tesla scanner(Magnetom Avanto,Siemens AG,Erlangen,Germany) using a dedicated receive only spine coil.After initial blinding,each dataset was evaluated by 2 board certified radiologist with more than 5 years experience in musculoskeletal imaging.In total 2364 facet joints were graded.Degenerative changes of the facet joints were evaluated according to the 4-point scale as proposed by Weishaupt et al Functional status was assessed using the ODI.The index is scored from 0 to 100 and interpreted as follows:0%-20%,minimaldisability; 20%-40%,moderate disability; 40%-60%,severe disability; 60%-80%,crippled; 80%-100%,patients are bedbound.Spearman's coefficient of rank correlation was used for statistical analysis,with significance set at P < 0.05.RESULTS:In total 2364 facet joints at lumbar levels L4/5 and L5/S1 were analysed in 591 individuals.FJOA was present in 97%(L4/L5) and 98%(L5/S1).At level L4/5(left/right) 17/15(2.9%/2.5%) were described as grade 0,146/147(24.7%/24.9%) as grade 1,290/302(49.1%/51.1%) as grade 2 and 138/127(23.4%/21.5%) as grade 3.At level L5/S1(left/right) 10/11(1.7%/1.9%) were described as grade 0,136/136(23.0%/23.0%) as grade 1,318/325(53.8%/55.0%) as grade 2 and 127/119(21.5%/20.1%) as grade 3.Regarding the ODI scores,patients' disability had a minimum of 0% and a maximum of 91.11% with an arithmetic mean of 32.77% ± 17.02%.The majority of patients(48.39%) had moderate functional disability(21%-40%).There was no significant correlation between FJOA and ODI on both sides of lumbar level L4/5 and on the left side of lumbar level L5/S1.A weak positive correlation was evaluated between ODI and FJOA on the right side of lumbar level L5/S1.CONCLUSION:The missing correlation of FJOA and ODI confirms our clinical experience that imaging alone is an insufficient approach explaining low back pain.Clinical correlation is imperative for an adequate diagnostic advance in patients with low back pain.

    • 摘要: AIM: To compare freeze-dried strawberry(FDS) beverage and strawberry-flavored drink effects on lipid profile and blood pressure in type 2 diabetic(T2D) patients.METHODS: In a randomized, double-blind, controlled trial, 36 subjects with T2D(23 females; mean ± SE age: 51.57 ± 10 years) were randomly divided into two groups. Participants consumed two cups of either pure FDS beverage(each cup containing 25 g freeze-dried strawberry powder equivalent to one serving of fresh strawberries; intervention group) or an iso-caloric drinkwith strawberry flavoring(similar to the FDS drink in fiber content and color; placebo group) daily for 6 wk. Anthropometric measurements, 3 d, 24 h dietary recall, and fasting blood samples were collected at baseline and at weeks 6 intervention. After lying down and relaxing for approximately 10 min, each participant's blood pressure was recorded in triplicate with 5 min intervals; recordings were made at baseline and the trial end-point. Each participant's lipid profile was assessed before and after intervention.RESULTS: Assessment at the weeks 6 intervention showed a significant reduction from baseline in total cholesterol levels and total cholesterol to high-density lipoprotein cholesterol(HDL-C) ratio in the intervention group(179.01 ± 31.86 to 165.9 ± 32.4 mg/L; P = 0.00 and 3.9 ± 0.88 to 3.6 ± 0.082 mg/L; P = 0.00 respectively), but the change was not significantly different between the two groups(P = 0.07, P = 0.29 respectively). Systolic blood pressure levels were significantly reduced from baseline in both the FDS and placebo drink groups(129.95 ± 14.9 to 114.3 ± 27.5 mm Hg; P = 0.02 and 127.6 ± 15.6 to 122.9 ± 14.47 mm Hg; P = 0.00 respectively), but the reduction was not significantly different between the two groups. Diastolic blood pressure was significantly reduced post-intervention in the FDS drink group compared to placebo group(78.7 ± 7.2 vs 84.4 ± 5.8; P = 0.01), the reduction was also significant within the FDS drink group(84.2 ± 8.03 to 78.7 ± 7.2; P = 0.00). Triglycerides, HDL-C concentrations and anthropometric indices showed no significant differences between or within groups. CONCLUSION: Short-term FDS supplementation improved selected cardiovascular risk factors in subjects with T2 D. Long-term effects on other metabolic biomarkers need to be investigated in future trials.
