为有效鉴别鲜茧生丝和干茧生丝,借助电子显微镜、傅里叶红外光谱、X射线衍射仪、热重分析仪和表面接触角测试等分析手段对鲜茧生丝和干茧生丝的表面形貌和结构、性能进行表征和分析,并基于它们结构与性能的差异建立了较为准确和简单易行的生丝鉴别方法.结果表明:2种生丝的化学组成相似,其大分子二级构象均主要为β折叠构象,且热分解曲线也基本一致;但与干茧生丝相比,鲜茧生丝的表面较为粗糙,茧丝之间空隙较大,结晶度略低且亲水性好.使用测定生丝束在十二烷基苯磺酸钠水溶液中沉降时间的方法能够有效地鉴别2种生丝,其中鉴别鲜茧生丝的准确率达到100%,而鉴别干茧生丝的准确率接近80%.

为探讨鲜茧前处理方式对生丝性能的影响,选择干茧、冰冻茧为研究对象,采用真空渗透、煮茧(不煮茧)、立缫工艺生产35 dtex(30/33D)生丝,按照织造要求进行浸泡处理,测试生丝的性能.结果表明:干茧生丝、鲜茧生丝的抱合性能良好,平均抱合次数均超过100次;浸泡处理后,鲜茧生丝丝胶稳定性较差,其抱合力下降幅度较大;鲜茧经过煮茧工艺,在一定程度上能够提高生丝的抱合稳定性;干茧生丝、鲜茧生丝及其浸泡后生丝的结晶度、取向度相差不大;干茧生丝、鲜茧生丝及其浸泡后生丝的断裂强度均超过3.5 cN/dtex,能满足基本的织造要求.%For exploring the effect of fresh cocoon pretreatment on the properties of raw silk , the dried cocoon and fresh cocoon were selected as the objects of study ,and vacuuming penetration ,cocoon cooking(non cocoon cooking) and multi-end silk reeling were applied to produce raw silk 35 dtex (30/33 D) .Then the raw silk was soaked according to weaving requirements and the performance of raw silk was tested .The experimental results show that dried cocoon (fresh cocoon) raw silks have good cohesion ,and the average times is more than 100 ;the cohesion of fresh cocoon decreases greatly and sericin stability is poor after soaking ;cocoon cooking can improve cohesion stability to certain degree .The crystallinity and orientation of these raw silks are close .The breaking strength of these raw silks are over 3 .5 cN/dtex ,so they can satisfy the basic requirement of weaving .

In order to better understand the changes of cocoon properties after cryopreservation of fresh cocoon under different freezing conditions,the cocoons from the same place were chosen as the study subjects and were preserved under different freezing conditions.Cocoon quality and cocoon layer permeability of dried cocoon,fresh cocoon and fresh cocoon with different freezing time at different temperatures were tested and analyzed.The results show that in the same process conditions,the moisture content of pupa decreases slightly after cryopreservation,and the moisture content of cocoon layer increases.The test results of good pupa rate show that if the pupa remains fresh for a long time,the storage temperature should be below -20 °C.After cryopreservation of fresh cocoon,the early sericin dissolution-loss ratio of cocoon layer becomes larger,the permeability of cocoon layer increases,the times of inner dropping end decreases,which is conducive to the smooth dissociation of bave in silk reeling process.%为了更好地了解鲜茧经不同冷冻条件保存后茧质性能变化,以同一庄口的蚕茧为研究对象,设置不同的冷冻条件进行保存.对干茧、鲜茧、不同温度冷冻不同时间后鲜茧的茧质以及茧层渗透性能进行测试和分析.结果表明:在其他工艺条件相同的条件下,鲜茧经冷冻保存后蛹体含水率略有减少,茧层含水率增大;好蛹率测试结果显示,如需较长时间保持蛹体新鲜,储藏温度至少要低于-20 °C;鲜茧经冷冻保存后茧层初期丝胶溶失率变大,茧层渗透性能增强,内层落绪次数减少,有利于缫丝过程中茧丝的顺利离解.

为对鲜茧生丝微量组分的提取方法进行探究,文章通过高效液相色谱(HPLC)研究了不同质量分数乙醇/水溶液的提取溶剂和提取条件(温度、时间和浴比)对鲜茧生丝多酚微量组分提取效率的影响,并采用正交试验优选出较佳提取条件.结果表明,不同溶剂鲜茧生丝提取物的HPLC图有明显差异,表现为乙醇质量分数越高,色谱峰强度越低,组分含量越低.不同提取条件对保留时间为10~16min组分含量均有影响,其中以温度的影响最为显著.同时研究表明,纯水是鲜茧生丝多酚微量组分最佳提取溶剂,以纯水为提取溶剂的较佳提取条件为:温度40°C,时间90min,浴比1;25.%To explore the extraction method of trace components in fresh raw silk, this paper is intended to study the effect of extraction solvents (ethanol/water solution) of different mass fractions and extraction conditions (temperature, time and liquor ratio) on extraction efficiency of polyphenols trace components in fresh raw silk via high performance liquid chromatography (HPLC), figure out the optimum extraction conditions via orthogonal test.The results indicate that the HPLC chromatograms of extracts from fresh raw silk with different solvents significantly differ.Specifically, with the mass fraction of ethanol is in negative correlation with both the intensity of chromatographic peaks and the content of components.All the extraction conditions have impact on the content of components having retained for 10 to 16min, and the impact of temperature is most significant.Meanwhile, researches show that pure water is the best extraction solvent of polyphenols trace components in fresh raw silk, of which the optimum extraction conditions are: temperature of 40°C, duration of 90min, and liquor ratio of 1:25 (g/mL).
