Phospholipids from apricot seed coat were extracted by using ultrasonic assisted extraction and then analyzed by high performance liquid chromatography. The phospholipids yield was served as the index, the ex-traction technology was investigated by single factor experiment and orthogonal experiment. The results indicat-ed that the optimal extraction parameters were as follows:the extraction solvent was hexane-ethanol (1:1, by volume), the material liquid ratio was 1:15 (g/mL), the ultrasonic power was 450 W, and the extraction time was 50 min, the total phospholipids content in apricot seed coat was 0.0912%under these conditions. Phos-pholipids were separated on a Silica gel column (ZORBAX Rx-SIL, 4.6 mm×250 mm, 5μm), with acetoni-trile-methanol (15:85, by volume) as the mobile phase at the flow rate of 1.0 mL/min, and detected by a UV detector. The column temperature was 35 °C, the injection volume was 20 μL, and the phosphatidyl ethanolamine(PE) content in seed coat was 0.3934 mg/g .%以山杏内种皮为原料,采用超声波辅助萃取法提取磷脂类化合物,并利用高效液相色谱法分离测定含量.以总磷脂得率为评价指标,通过单因素试验和正交试验优化超声波辅助萃取条件.结果表明,以正己烷-乙醇(1:1,体积比)为提取溶剂,料液比为1:15(g/mL),超声功率为450 W,超声时间为50 min最优,在此条件下,山杏内种皮总磷脂含量为0.0912%.高效液相色谱-紫外检测器法测定磷脂含量,采用ZORBAX Rx-SIL型硅胶柱(4.6 mm×250 mm,5μm),流动相为乙腈-甲醇(15:85,体积比),流速1.0 mL/min,柱温35°C,进样量20μL,测得山杏内种皮中磷脂酰乙醇胺(PE)含量为0.3934 mg/g.
