摘要:
为探讨羊栖菜多糖对果蝇抗氧化作用和寿命的影响, 采用黑腹果蝇为研究对象, 添加羊栖菜多糖低、中、高剂量组 (质量浓度分别为1.0%, 2.0%, 4.0%) 的培养基饲喂, 每3 d更换1次培养基, 35日龄后, 分别测定果蝇组织匀浆中T-AOC、ASAFR、CAT、SOD的活性及MDA含量, 并每日定时观察记录果蝇存活数, 统计平均寿命、最高寿命、半数死亡期, 计算平均寿命延长率和最高寿命延长率.结果表明经过各剂量组的羊栖菜多糖饲喂后, 均可增强黑腹果蝇组织匀浆中T-AOC、ASAFR、CAT、SOD活性, 其中2.0%剂量组效果最佳, 分别提高雌性26.22%, 21.51%, 23.72%, 31.30%, 雄性18.50%, 23.07%, 15.75%, 24.85% (P<0.05), 降低果蝇体内MDA含量, 雌性12.78%, 雄性20.06% (P<0.05).不同羊栖菜多糖剂量组均能延长果蝇的生活寿命, 其中4.0%剂量组显著延长雌雄果蝇的最高寿命至50, 48 d (P<0.05), 提高雌雄果蝇的平均寿命达7 d和10 d (P<0.05).羊栖菜多糖抗氧化作用较强, 该结果为羊栖菜多糖应用于保健品和化妆品领域的开发提供了一定的实验基础.%In order to explore the effect of the Sargassum fusiforme polysaccharides on the anti-oxidation action and longevity of the fruit flies, the drosophila melanogaster is taken as study objects, which are fed by the growth mediums added with the Sargassum fusiforme polysaccharides that are divided into low, medium and high dose groups (the mass concentrations are 1.0%, 2.0%, 4.0%respectively), the growth mediums shall be replaced every 3 days, when the fruit flies are 35 days old.The activity of the T-AOC, ASAFR, CAT, SOD and the MDA content in the tissue homogenate of the fruit flies shall be determined respectively.The survival number of the fruit flies shall be recorded.The average life span, the maximum life span and the half decline phase shall be summarized, and the average life span extension rate and the maximum life span extension rate shall be calculated in a daily manner.The results showed that after the feeding of the Sargassum fusiforme polysaccharides in each dose group, the activity of the T-AOC, ASAFR, CAT, SOD in the tissue homogenate of the drosophila melanogaster are all enhanced and among them, the 2.0% dose group had the best effect, which are increased by 26.22%, 21.51%, 23.72%, 31.30% respectively in the female flies, by 18.50%, 23.07%, 15.75%, 24.85% (P < 0.05) in the male flies, and the MDA content in the fruit flies are reduced by 12.78% and20.06% (P < 0.05) in the female flies and male flies respectively.All the three types of dose groups can prolong the life span of the fruit flies, among which the 4.0% dose group is significantly prolonged: the maximum life span of the female and male fruit flies to50 d, 48 d (P < 0.05) respectively, and is increased the average life span of the female and male fruit flies by 7 d and 10 d (P < 0.05) respectively.The anti-oxidation effect of the Sargassum fusiforme polysaccharides is strong, which provides an experimental basis for the application of the Sargassum fusiforme polysaccharides in the health products and cosmetics.
