    • 摘要: 一、首次现场4K制作2015江苏卫视跨年演唱会于2014年12月31日在南京奧体中心向全国进行高清直播。与此同时,江苏台技术人员在场馆功能房里搭建了一套4讯道4K EFP系统开始探索和尝试超高清电视节目制作,该系统配备了1套Sony MVS70004 K切换台、4套Sony HDCU 2080+BPU4000基站、4套Sony PVM F55+CA4000摄像机、Sony PVM X300监视器和1套Sony PWS4400服务器等设备,参与到2015江苏卫视跨年演唱会的现场制作,这是江苏省广播电视总台首次尝试超高清电视节目制作,算是摸着石头过河。

    • 摘要: 测定马铃薯M病毒(Potato virus M,PVM)寄主范围,并作PVM接种条件优化试验.结果表明,PVM仅侵染15种指示植物中的4种,各种植物症状表现不同.苋色黎(Chenopodium amaranticolor)和千日红(Gomphrena globosa L.)接种叶片上产生枯斑,可作为PVM枯斑寄主;36号烟(Nicotiana occidentalis“pi”)系统发病,新生叶片产生皱缩、泡斑、叶边缘卷曲等症状,植株长势矮小,可作为PVM系统侵染鉴定寄主;PVM系统侵染番茄(Lycopersicon pimpinellifolium)速度快,但无症状,番茄可作为PVM繁殖寄主.PVM虽可侵染不同生育期寄主,以接种中上部叶位侵染效果最好,但在不同寄主不同叶位分布并不一致,番茄体内中部叶片病毒含量相对较高,在苋色黎上则分布较均匀.明确PVM最适培养温度为23 ~29°C.最高稀释度64倍时(OD405=0.201)仍可成功侵染苋色黎.为PVM纯毒源扩繁及制备该病毒抗血清提供基础数据.%In this study,the host range of potato virus M (PVM) was determined,and on the basis of this,PVM inoculation conditions were optimized.The results showed that PVM only infected four indicator plants among the 15 species tested,and the symptoms were different.On the inoculated leaves of Chenopodium amaranticolor and Gomphrena globosa L.expressed lesion,so they could be used as the lesion host of PVM.The Nicotiana occidentalis "pi" expressed systemic symptom,on the new leaves of which expressed crinkling,bubble spot,leaf curl and dwarf,so it could be used as the identification host of systemic infection.Lycopersicon pimpinellifolium could be systemically infected.The speed was fast,but did not show symptoms,so it could be used as the breeding host of PVM.Although PVM could infect the host plants of different growth stages,the leaves at mid-upper positions got the best infection result.However,the concentration of PVM was inconsistent between the leaves of different positions,with the virus content in leaves being relatively high in the middle part of the Lycopersicon pimpinellifolium,while the distribution in Chenopodium amaranticolor being even.The optimal culture temperature of PVM was 23-29 °C.The highest dilution of 64 times (OD405=0.201) could still successfully infest Chenopodium amaranticolor.These results may provide the basic data for the preparation of the virus antiserum by the expansion of pure PVM.

    • 摘要: 目的:观察PVM新辅助化疗(NACT)方案治疗玉b-Ⅱb期宫颈癌的近期效果。方法用PVM(顺铂联合长春新碱及丝裂霉素)化疗方案对40例玉b-Ⅱb期宫颈癌进行2~3个疗程的术前静脉化疗,观察近期疗效及宫颈旁浸润、静脉癌栓、淋巴转移、毒副反映等情况。结果40例宫颈癌患者经过术前化疗,宫颈出血明显减少,局部肿瘤明显缩小,宫旁组织变软,使手术顺利进行。其中1例为低分化腺癌,化疗后无变化,给予放射治疗。结论采用PVM宫颈癌新辅助化疗可以缩小肿瘤体积,降低肿瘤的分期,提高手术的治疗效果。

    • 摘要: Potato virus X (PVX), potato virus M (PVM) and potato virus A (PVA) are important viruses which can cause potato seed degeneration and sometimes complex infection. Therefore, it is very important to establish a fast and exact detection system. In this research, specific primer for each virus was identified by reading both Chinese and English literatures, and evaluation and selection. Then, a stable RT-PCR reaction system was established through optimized density of Mg2+, dNTPs and Taq DNA polymerase and the expected fragments of 711 bp (PVX), 520 bp (PVM) and 273 bp (PVA) were successful y amplified. This system and DAS-ELISA method were simultaneously used to detect 150 potato field samples, and the results were the same. The triplex RT-PCR system is more sensitive and specific, and could be used to detect potato field samples.%马铃薯X病毒(PVX)、马铃薯M病毒(PVM)和马铃薯A病毒(PVA)是导致马铃薯种薯退化的重要病毒,有时复合侵染,因此建立快速、准确检测体系尤为重要。试验从中、英文文献中查找引物,通过筛选和综合评价,每种病毒确定1对特异性引物,再通过对PCR部分Mg2+、dNTPs、TaqDNA聚合酶浓度梯度优化,最终建立了稳定的三重RT-PCR反应体系,得到长度分别为711、520、273 bp的特异性条带。应用该体系和DAS-ELISA方法同时对田间150份马铃薯毒源样品进行检测,两种方法检测结果吻合,三重RT-PCR体系的灵敏度更高、特异性更强,可以用于马铃薯田间样品检测。

    • 摘要: 马铃薯X病毒(PVX)、马铃薯Y病毒(PVY)、马铃薯S病毒(PVS)、马铃薯卷叶病毒(PLRV)、马铃薯M病毒(PVM)和马铃薯A病毒(PVA)是马铃薯生产田中发生率较高、危害较严重的病毒,因此,建立特异性强、灵敏度高、检测速度快的RT-PCR分子检测体系对保障马铃薯种薯质量具有非常重要的意义。试验筛选了6种病毒的特异引物、优化了PCR部分的试剂以及确定了退火温度。结果表明,当Mg2+浓度为2.6 mmol/L、dNTPs浓度为0.1 mmol/L、Taq DNA聚合酶浓度为0.03 U/μL,以及退火温度为55.5°C时反应体系最稳定。最终,建立了一套通用RT-PCR检测体系,只需要变换每种病毒的引物,就可以实现对PVX、PVY、PVS、PLRV、PVM和PVA病毒的RT-PCR检测。每种病毒检测灵敏度均能达到pg以上。%The higher incidence and more serious viruses in potato fields are potato virus X (PVX), potato virus Y (PVY), potato virus S (PVS), potato leafroll virus (PLRV), potato virus M (PVM), and potato virus A (PVA), so it is very important to guarantee the quality of seed potato by establishment of RT-PCR molecular detection system with specificity, high sensitivity and fast detection speed for these viruses. In this research, the primer for each virus was screened, PCR reagents were optimized, and annealing temperature was determined. The results indicated that when the concentration of Mg2 + was 2.6 mmol/L, dNTPs was 0.1 mmol/L, Taq DNA polymerase was 0.03 U/μL, and annealing temperature was 55.5°C, the reaction system was the most stable. Finally, a universal RT-PCR detection system was established, which only need to change primer for each virus when the RT-PCR detection for PVX, PVY, PVS, PLRV, PVM and PVA was needed. The detection sensitivity of each virus could reach the level of pg.
