A method for constructing a non-de novo synthetic high-lipid-yielding recombinant Mucor circinelloides bacterium, a recombinant bacterium constructed by the same, and an application thereof, pertaining to the field of genetic engineering. The method for constructing a non-de novo synthetic high-lipid-yielding recombinant Mucor circinelloides bacterium comprises: ligating a nucleotide sequence of diacylglycerol acyltransferase (DGAT) to an integrated plasmid pMAT2075, electro-transforming the recombinant plasmid into the protoplast of a uracil-deficient Mucor circinelloides bacterium, screening for a positive clone, and overexpressing the DGAT in the uracil-deficient Mucor circinelloides bacterium to obtain the non-de novo synthetic high-lipid-yielding recombinant Mucor circinelloides bacterium. Compared with a control strain Mc2075, the lipid content of the constructed recombinant strain Mc-DGAT in a medium supplemented with soybean oil showed greater change, and a lipid content of 53% could be obtained in the expression strain Mc-DGAT.
